Abstract:
Professor Ding and his team have been promoting the quality and quantity of pluripotent stem cells and its clinical and translational application based on the study of the epigenetic and 3D chromatin regulatory mechanism of pluripotency, and the main achievements include: (1) discovered the function and mechanism of DNA demethylase TET1 and TET2 in promoting the efficiency and quality of iPSC generation for the first time (Nature, 2013); (2) further discovered a novel pluripotency factor, TEX10, which recruits epigenetic modifiers to super-enhancer regions and promote stem cell pluripotency (Cell Stem Cell, 2015); (3) elucidated the role of YY1 in regulating epigenetic modification of super-enhancers through its interaction with the BAF complex to promote gene expression of pluripotency based on the previous study (Stem Cell Reports, 2018); and (4) optimized the traditional methods of proteomics for above epigenetic studies (Cell Research, 2012). These studies provide novel epigenetic pathways to promote the quality and quantity of pluripotent stem cells, and provided a theoretical basis for its further clinical application. To further promote the quality and quantity of pluripotent stem cells, the team studies the functions of super-enhancers and its mechanisms in 3D chromatin levels based on epigenetic regulation for pluripotency regulation. They further found the key reprogramming factor can facilitate super-enhancer mediated 4D nucleome by phase separation to induce reprogramming.
